چکیده

THE ELECTROPHYSIOLOGIC EFFECTS of adenosine and other adenine derivatives were first described by Drury and Szent-Gyorgyi in 1929.' These investigators clearly demonstrated that intravenous administration of adenosine produced a decrease in sinus rate and transient atrioventricular (AV) block. Adenosine is a mediator of many physiologic phenomena and most of the interest in its effects on the cardiovascular system have, until recently, been centered on its ability to regulate regional blood flow in the heart and other organs. During the last several years, we and several other investigators have reexamined the electrophysiologic effects of adenosine.2'6 In our initial experiments, we showed that adenosine produced variable degrees of dose-related reversible AV block in guinea pig and rabbit hearts perfused by the Langendorff technique. The negative dromotropic effect was localized to AV nodal tissue.5 6 In the anesthetized dog, perfusion of the sinus and AV nodal arteries with solutions containing adenosine produced a decrease in sinus nodal discharge and an increase in AV nodal conduction time, respectively.3'4' 7,8 Studies in isolated segments of the AV node and the isolated perfused mammalian heart have confirmed the earlier observations that adenosine impairs conduction through AV nodal tissue, and that its action is restricted to the proximal portion of the node, i.e., the slow channel-dependent tissue of the AV node.9 In addition to these effects on the sinoatrial and AV nodes, adenosine shortens the action potential duration and hyperpolarizes the membrane of atrial cells. Background: The 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase inhi-bitors (statins) have revolutionized the treatment of hypercholesterolemia. Some evide-nce indicated the role of nodal refractoriness and concealed conduction in anticipating the ventricular rate during atrial fibrillation. Recent evidence has indicated that statins can reduce the incidence of both supraventricular a...

Effect of adenosine on atrioventricular conduction. I: Site and characterization of adenosine action in the guinea pig atrioventricular node.

Adenosine has a negative dromotropic effect and modulates hypoxia-induced atrioventricular (AV) conduction delay. To further characterize the negative dromotropic effect of adenosine in the guinea pig heart, we determined the site of adenosine-induced AV conduction block; the effect of uptake and deamination of adenosine on its concentration-negative dromotropic effect, and the adenosine recept...

Modulation of extracellular atrioventricular node field potential pattern and ventricular rhythm by morphine in experimental atrial fibrillation in isolated rabbit heart

Introduction: Endorphins are produced by cardiomyocytes, and exert different effects on the heart. The aim of the present study is to assess morphine effects on extracellular atrioventricular (AV) node field potential pattern and ventricular rhythm of isolated rabbit heart during experimental atrial fibrillation (AF). Methods: Effects of different concentrations of morphine (10, 20, 50 and 1...

Effect of adenosine on atrioventricular conduction. II: Modulation of atrioventricular node transmission by adenosine in hypoxic isolated guinea pig hearts.

Adenosine as well as hypoxia and ischemia are known to cause atrioventricular conduction block. To test the hypothesis that adenosine is the primary mediator of hypoxia-induced atrioventricular conduction delay in isolated perfused guinea pig hearts, we characterized a) the time courses of hypoxia-induced adenosine release and delay in atrioventricular conduction, b) the relationships between o...

AVNRT, (Atrioventricular nodal reentry tachycardia), atrial tachycardia and atrial flutter are 3 kinds of supraventricular tachycardia, which their mechanism are explained based on reentry. A 60-years-old man is presented with all of the above-mentioned arrhythmias, responsive to intravenous injection of adenosine. Radiofrequency ablation of the slow pathway territories cured all of them. There...
